        教學簡介:

        本科生課程《蛋白質(zhì)化學與工程》,研究生課程《蛋白質(zhì)結(jié)構與現(xiàn)代藥物開發(fā)》。指導學生獲得福建省研究生優(yōu)秀學位論文。

        科研簡介:

        要從事蛋白的結(jié)構和功能研究,并基于結(jié)構進行多肽和蛋白類藥物的改造和評價。目前已發(fā)表SCI論文九十余篇,先后主持國家自然科學基金面上項目及福建省自然科學基金重點項目等。主要研究領域包括:

        1、腫瘤遷移相關蛋白的結(jié)構和功能研究:幾乎90%癌癥病人的死亡是由于癌細胞的轉(zhuǎn)移性擴散引起。目前針對原發(fā)性腫瘤的治療卓有成效,但靶向腫瘤轉(zhuǎn)移方面的進展卻不盡人意。我們主要針對腫瘤遷移相關的一些蛋白如尿激酶受體系統(tǒng)和多種絲氨酸蛋白酶,及其與抑制劑的復合物進行X-射線晶體結(jié)構研究,并針對這些結(jié)構指導抑制劑和靶向彈頭的改造,以發(fā)展高效特異的腫瘤治療藥物。

        2、血栓形成相關蛋白的結(jié)構和功能研究:人體保持血液處于正常流動狀態(tài),主要依賴于凝血系統(tǒng)和纖溶系統(tǒng)處于一個動態(tài)平衡。病理條件下兩個系統(tǒng)的功能異常將導致出血或血栓形成。我們針對凝血和纖溶相關的一些蛋白如纖溶酶原激活物tPA、多種凝血因子及它們的調(diào)控蛋白PDI進行結(jié)構和功能的研究,并基于這些結(jié)構進行蛋白質(zhì)工程蛋白及抑制劑的設計和改造,以發(fā)展高效且無出血副作用的溶栓和抗血栓形成藥物。

        3、光動力抗腫瘤和抗菌研究:光敏劑在特定波長的光激發(fā)下可產(chǎn)生具有生物毒性的單線態(tài)氧或者自由基,進而殺死細胞或微生物,可用于抗腫瘤和抗菌方面的治療。傳統(tǒng)的光敏劑存在靶向性差,水溶性不佳等問題。我們對光敏劑進行一系列的修飾和包裹,發(fā)展高靶向性光敏劑,用于抗腫瘤和耐藥菌的治療。
