        科研簡(jiǎn)介:

        研究方向主要集中在抑癌基因基因p53和腫瘤代謝重編程方面。抑癌基因p53在維持基因組穩(wěn)定以及抑制腫瘤發(fā)生方面有著重要作用。p53是腫瘤中突變頻率最高的基因,超過(guò)50%的腫瘤包含p53突變,80%以上的腫瘤中p53信號(hào)通路功能異常。作為一個(gè)轉(zhuǎn)錄因子,p53蛋白能被細(xì)胞內(nèi)外各種干擾,包括DNA損傷、缺氧、核糖體合成受阻乃至原癌基因激活等激活,從而進(jìn)一步選擇性的激活其下游的各種目的基因,引起諸如細(xì)胞凋亡、細(xì)胞周期阻滯以及細(xì)胞衰亡等過(guò)程,避免細(xì)胞癌化。代表性成果包括:

        1)發(fā)現(xiàn)了兩個(gè)對(duì)腫瘤代謝有重要影響的p53目的基因GLS2和Parkin (PNAS, 2010& 2011, IF:9.504),并在此基礎(chǔ)上進(jìn)一步分析了它們?cè)谀[瘤的作用和分子機(jī)制,闡明了(a) GLS2通過(guò)與Rho GTP酶Rac1的結(jié)合降低Rac1活性,抑制由Rac1活性升高引起的腫瘤生長(zhǎng)與轉(zhuǎn)移 (Elife, 2016,IF: 7.616);(b) Parkin作為一個(gè)E3泛素連接酶,能夠結(jié)合并機(jī)制降解HIF-1α,從而抑制由HIF-1α引起的腫瘤生長(zhǎng)與轉(zhuǎn)移為相關(guān)腫瘤的治療提供了新的策略(Nature Communication, 2017, IF: 12.353)。

        2)首次報(bào)道了腫瘤相關(guān)p53突變能夠通過(guò)改變RhoA活性促進(jìn)葡萄糖轉(zhuǎn)運(yùn)蛋白GLUT1向細(xì)胞膜上轉(zhuǎn)移,從而增加葡萄糖的吸收和糖酵解,進(jìn)而促進(jìn)腫瘤生長(zhǎng),為含有p53突變的腫瘤治療提供了新思路(Nature Communication, 2013, IF: 12.353)。

        3) 抑癌基因CUL3能夠與ATP-檸檬酸裂解酶(ACLY)結(jié)合,導(dǎo)致其泛素化降解,從而降低細(xì)胞脂類合成水平,抑制腫瘤的生長(zhǎng)。因此,在CUL3突變或缺失的肺癌中,使用ACLY抑制劑能夠有效的減緩肺癌的生長(zhǎng) (Gene & Development, 2016, IF: 9.462)。
